【摘要】:Thisinvivoprocedureisdesignedtotesttheabilityofhygienichandwashandhandrubagentstoreducelevelsofselectedinfectiousvirusesfromexperimentallycontaminatedfingerpadsofadults.Sincethetwothumbpadsandalleightfingerpadscanbeusedinanygiventest,itallowsfortheincorporationofinputviruscontrol(two),virusremainingviableaftertheinoculumhasbeenallowedtodry(two),viruseliminatedaftertreatmentwithacontrolorreferencesolution(two),anduptofourreplicatestoassessthevirus-eliminatingefficiencyofthesubstanceundertest.Nomorethan100x03BC;Lofthevirussuspensionarerequiredtocompleteonetest.Theresultsoftestingwiththistestmethodmayformthebasisforfurthertestsusingasuitablewhole-handtestprotocol(forexample,TestMethodE2011).Thistestmethodisdesignedtobeperformedbyatrainedindividual,whoisresponsibleforchoosingtheappropriatehostsystemforthetestvirusandapplyingthetechniquesnecessaryforpropagationandmaintenanceofhostandtestvirus.Forareferencetext,refertoLennetteetal(8).Whereasthemethoddescribedhererelatestotestingwithvirusesofhumanorigin,itcanbereadilyadaptedtoworkwithanimalpathogenicvirusesaswellasbacteriophages.Standardmethodsforworkingwithbacteria(TestMethodE2276)andfungi(TestMethodE2613)arealsoavailable.Infectiousmicroorganismsleftonhandsafterwashingcanbereducedfurtherbydryingthewashedhandswithpaper,cloth,orwarmair(9).Astepforthedryingoffingerpadsafterexposuretothecontrolortestsubstance,therefore,hasnotbeenincludedtoavoidvirusremovalbythedryingprocessitself.Thistestmethodisnotmeantforusewithsurgicalhandscrubsorpreoperativeskinpreps.Thelevelofviablevirusoneachfingerpadafterthedryingoftheinoculumshouldnotbelessthan104infectiousunitswhichwouldpermitthedetectionofuptoa4log10reductionintheinfectivitytiterofthevirusbythetestsubstanceundertheconditionsofthistestmethod.1.1Humanskinisnotknowntocarryvirusesasapartofitsresidentmicrobiota.Handstransientlycontaminatedwithvirusescan,however,actasvehiclesforthespreadofmanytypesofviralinfections.Handhygieneismeanttoreducetheloadofvirusesandothertransientmicroorganismsonhands,therebyreducingtheriskofdiseasetransmission.Suchreductionsinthevirusloadmaybeduetoacombinationofvirusinactivationandmechanicalremovalofinfectiousvirusfromtheskin.1.2Thistestmethodisdesignedtodeterminethecomparativevirus-eliminatingeffectivenessofmicrobicidalornon-microbicidalformulations.Thistestmethodisnotmeantforusewithsurgicalhandscrubsorpreoperativeskinpreps.Note18212;Thetestmethodshouldbeperformedbypersonswithtraininginvirologyinfacilitiesdesignedandequippedforworkwithinfectiousagentsatbiosafetylevel2(6).1.3ThevaluesstatedinSIunitsaretoberegardedasstandard.Nootherunitsofmeasurementareincludedinthisstandard.1.4Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddet......

适用范围
本标准包括三个方法：第一法高效液相色谱法，第二法液相色谱-质谱/质谱法，第三法气相色谱-质谱联用法，检测时，应根据检测对象及其限量的规定，选用与其相适应的检测方法。
本标准规定了原料乳、乳制品以及含乳制品中三聚氰胺的三种测定方法，即高效液相色谱法（HPLC）、液相色谱-质谱/质谱法（LC-MS/MS）和气相色谱-质谱联用法[包括气相色谱-质谱法（GC-MS），气相色谱-质谱/质谱法（GC-MS/MS）]。
本标准适用于原料乳、乳制品以及含乳制品中三聚氰胺的定量测定；液相色谱-质谱/质谱法、气相色谱-质谱（包括气相色谱-质谱/质谱）法同时适用于原料乳、乳制品以及含乳制品中三聚氰胺的定性确证。
本标准高效液相色谱法的定量限为2 mg/kg，液相色谱-质谱/质谱法的定量限为0.01 mg/kg，气相色谱-质谱法的定量限为0.05 mg/kg（其中气相色谱-质谱/质谱法的定量限为0.005 mg/kg）。
